Darkfield microscopy is a lighting method in microscopy where the specimen for examination is illuminated from the sides rather than from directly below or from directly above. The technique gives a black background with a brightly light specimen. Darkfield light microscopes can be either low power stereoscopic microscopes or high power compound light microscopes. The low power models are useful for gemology and gemstone grading. They consist of a darkfield light stereo microscope combined with a darkfield condenser and a jewelry clip for holding the gemstone. High power compound light microscopes can be fitted with a darkfield kit to provide a darkfield microscopy solution. The normal brightfield abbe condenser is changed to a darkfield condenser. This provides darkfield light on the specimen contained on a microscope slide.

The most common application for darkfield microscopy in high power applications is for use with live blood cell analysis. The drop of blood is placed on the blank microscope slide, and a thin microscope cover slip is dropped on top of the live blood. Using this darkfield technique, features in the blood normally invisible in brightfield or phase contrast illumination become brightly illuminated and visible to the microscopist.

Live blood cell analysis is a whole area of alternative medicine based on analyzing the contents and condition of the live blood. Normally, blood is stained to allow better contrast. However, this staining also affects the natural balance of the blood cells, so what is viewed under stained blood is not exactly what takes place naturally inside the bloodstream.
